课题组的长期研究方向为炎症机制及干预研究。研究最终目标是阐明炎症相关细胞(如中性粒细胞)和因子在炎症的发生、发展和消退中的作用及规律,并研究和开发靶向干预的药物。课题组近10多年主要围绕中性粒细胞的死亡、功能及调控机制开展研究,主要研究进展如下:1. 发现中性粒细胞中丝氨酸蛋白酶3(proteinase 3,Pr3)能通过切割活化caspase 3来诱导中性粒细胞凋亡的新的分子机制;2. 六磷酸肌醇激酶-1 (Ip6k1)基因敲除或用TNP抑制IP6K1活性均能够有效的增强宿主对细菌的杀伤能力,但同时减少肺部中性粒细胞的招募进而降低肺部损伤。3.筛选到一个能在体外和动物体内高效抑制肿瘤坏死因子功能的全新小分子化合物C87。目前,课题组一方面将继续和深入上述研究,另一方面将开展炎症与自身免疫性疾病的关系以及髓系细胞与肿瘤的发展与转移的关系研究。
