实验室结合单细胞多组学和生物信息血方法,以血液系统为模型,研究表观遗传及基因组遗传多样性的复杂调控网络。主要方向包括:
造血调控网络:经典的造血分化模型表现为层级分化模式,随着今年来单细胞等技术的发展,这一经典模型受到了挑战并不断更新。如从造血干细胞到巨核细胞的旁路分化,以及造血干祖细胞间的连续分化状态。这其中,我们对各类细胞的身份、分化过程中命运的选择和分化路径等问题的理解仍不全面。为了回答这些问题,我们实验室通过整合单细胞多组学方法(包括基因组、转录组和表观组,如DNA甲基化和染色质可及性)来研究单个血细胞,这包含造血干细胞、祖细胞以及成熟的各谱系血细胞。我们也关注非正常造血和恶性血液病,希望为临床的血液病诊断和治疗提供新的方案。
生物信息方法开发:随着大量测序数据的积累,如何从大数据中准确且有效的挖掘出生物学机制,成为目前基于组学研究的主要挑战之一。因此,我实验室也致力与开发生物信息方法来推动组学数据分析。
