造血干细胞发育受到多种因素的精密调控,调控失衡将导致血液发育缺陷或相关疾病。深入系统地研究造血干细胞(HSC)发育中的关键因子及作用机制,对完善调控网络和治疗相关疾病具有重要意义。我们致力于揭示造血干细胞产生的分子调控机制,从细胞内源因子(遗传与表观遗传)和微环境角度,应用遗传学分析、高通量测序以及谱系示踪等技术,系统解析胚胎期造血干细胞发育的调控机制,包括:表观遗传修饰DNA甲基化及m6A甲基化影响造血干细胞命运决定(Dev, 2022; Nature, 2017);发现造血干细胞异质性起源(Cell Res, 2023);揭示RNA结合蛋白Csde1调控造血干细胞产生过程的机制(Dev, 2023);在造血干细胞分化过程中,Rac2调控T淋巴前体细胞迁移以及分化(Stem Cell Reports, 2020; J Immunol, 2020);发现表观遗传修饰DNA甲基化和RNA甲基化调控胚胎早期发育(EMBO J, 2017; Mol Cell, 2019)。
